Glycolipid Presentation by CD1
نویسنده
چکیده
T cells recognize lipid antigens as complexes associated with CD1 antigen-presenting molecules. The biophysical properties of lipids impose unique mechanisms for lipid delivery, cell internalization, membrane trafficking, processing of large glycolipid antigens and loadingofCD1molecules. T cells specific for lipid antigens participate in the regulation of immune response, protection during infections and pathogenesis of autoimmune diseases.

Glycolipid targets of CD1-mediated T-cell responses.
Members of the CD1 family of antigen-presenting molecules bind and present a variety of mammalian and microbial glycolipids for specific recognition by T cells. CD1 proteins accomplish their antigen-presenting function by binding the alkyl chains of the antigens within a deep, hydrophobic groove on the membrane distal surface of CD1, making the hydrophilic elements of the antigen available for ...
متن کاملInhibition of glycolipid shedding rescues recognition of a CD1+ T cell lymphoma by natural killer T (NKT) cells.
Neoplastic transformation of cells is accompanied by an aberration of cell surface glycolipid composition. These tumor-associated, altered glycosphingolipids are often shed into the tumor cell microenvironment and mediate immunosuppressive activity. The nature and form of glycolipids shed by a variety of tumor cell lines and the mechanism(s) of shedding have been well characterized. The murine ...
متن کاملCD1 and lipid antigens: intracellular pathways for antigen presentation.
Recently, different members of the CD1 family of MHC-like molecules have been shown to sample different intracellular compartments to present lipid and glycolipid antigens to T cells. Emerging models suggest that CD1 may have evolved to monitor the integrity of membrane lipids and/or to present microbial lipid antigens to both alpha beta and gamma delta T cells.

Five CD1 molecules are expressed in humans and it is unclear whether they have specialized or redundant functions. We found that sulfatide is a promiscuous CD1-binding ligand and have isolated T cell clones that are specific for sulfatide and restricted by distinct CD1 molecules. These clones have been used to compare the capacity of different CD1 to present the same glycolipid, to induce effec...

The reactivity of a group of mouse Valpha14+ NK T cell hybridomas was tested with a panel of analogs of the glycolipid alpha-galactosylceramide (alpha-GalCer). Interestingly, the nearly complete truncation of the acyl chain from 24 to 2 carbons does not significantly affect the mouse NK T cell response to glycolipid presented by either mouse CD1 (mCD1) or its human homolog CD1d (hCD1d). Therefo...
